Substances from African medicinal plants could help stop tumor growth

June 10, 2013 ? African medicinal plants contain chemicals that may be able to stop the spread of cancer cells. This is the conclusion of researchers following laboratory experiments conducted at Johannes Gutenberg University Mainz (JGU). The plant materials will now undergo further analysis in order to evaluate their therapeutic potential.

"The active substances present in African medicinal plants may be capable of killing off tumor cells that are resistant to more than one drug. They thus represent an excellent starting point for the development of new therapeutic treatments for cancers that do not respond to conventional chemotherapy regimens," explained Professor Thomas Efferth of the Institute of Pharmaceutical Sciences and Biochemistry -- Therapeutic Life Sciences at Mainz University. For the past four years, Efferth and biochemist Dr. Victor Keute of the University of Dschang in Cameroon have been studying the active substances in African plants such as the giant globe thistle, wild pepper, speargrass, and Ethiopian pepper.

Multi-drug resistance is one of the most feared problems in cancer therapy because in such cases most of the standard chemical cancer drugs used in therapy fail and the patient's chance of survival is thus dramatically reduced. The problem cannot usually be resolved by simply increasing the dosage as this also results in the exacerbation of undesirable side effects. "We are now looking for substances that can both break down tumor resistance and not produce side effects," continued Efferth, who also works with medicinal plants used in traditional Chinese medicine.

Many plants contain toxic substances that they use to protect themselves against predators and microbial diseases. Over the course of millions of years during which life has evolved on earth plants have managed to appropriate certain molecules to help them to offset their main disadvantage in the face of their enemies, i.e., their immobility and lack of an immune system. The challenge for the pharmacologists is now to determine which plant substances are medicinal and which are simply poisonous and dangerous.

During the joint project with Mainz, the Cameroonian scientist Victor Kuete has examined more than 100 spices and plants from his homeland for their cytotoxic effects on cancer cells. Awarded a Humboldt Research Fellowship, he can now continue and extend his investigations as a member of Thomas Efferth's work group in Mainz. "We have already found an entire series of benzophenones and other phytochemicals that are able to elude resistance mechanisms and thus offer many new opportunities for continued research," said Efferth.

The researchers are focusing on three different resistance mechanisms. Transporter-mediated resistance prevents drugs taking effect because a substance called P-glycoprotein promotes their efflux from cancer cells. In the case of tumor-suppressor-gene-mediated resistance, a mutation in protein p53 means that the cancer cells do not die but are resistant and become increasingly aggressive. Lastly, in oncogene-mediated resistance, the epidermal growth factor receptor (EGFR) sends signals into the interior of cells causing tumors to grow faster. The researchers in Mainz have cellular models of all three resistance mechanisms that will enable them to appropriately test the effectiveness of the substances obtained from plants.

In their latest of a total of eight publications produced to date, the research team reports that four naturally occurring benzophenones can prevent the proliferation of the tested cancer cell lines, including multi-drug resistant strains. "The benzophenones investigated are potentially cytotoxic substances that need to be more extensively investigated with the aim of developing new cancer drugs that are effective against susceptible and resistant cancers," claims the article recently published in the scientific journal Phytomedicine.

U.N. mediator sees breakthrough in Guinea election talks

CONAKRY (Reuters) - The Guinean government and opposition parties in the West African nation have made a breakthrough during talks on Sunday that could end a violent political impasse and pave the way for legislative elections, a United Nations envoy said.

President Alpha Conde's government and Guinea's opposition parties are locked in a United Nations-mediated talk over the organization of a long-delayed legislative election.

More than 50 people have been killed in three months of rallies by activists who accuse Conde of preparing to rig the poll, scheduled for June 30, in the world's largest bauxite exporter.

Said Djinnit, a U.N. envoy mediating the talks between government and opposition in the coastal capital of Conakry, said the parties have made significant progress over their demands and there were reasons for hope.

Djinnit said in return for some guarantees, Guinea's opposition parties have agreed to rejoin the electoral process, dropping a demand that South African company Waymark, charged with updating the voter register, be replaced.

The opposition accused the company of stuffing the electoral roll with the names of Conde's ethnic Malinke supporters, charges the company denied.

The opposition had also called for Guineans living overseas to be allowed to vote.

"Regarding the vote of Guineans abroad, the presidential camp, which had reservations on the issue, have lifted their opposition. It was agreed that Guineans living abroad could participate in elections," Djinnit said.

Decisions from the talks, if concluded, could affect the date of the election, he said.

Guinea's government was not immediately available for comment, but a spokesman for the opposition told Reuters that a minimum consensus had been reached and they were waiting for concrete actions from the government and Guinea's electoral commission.

"We have reasons to be cautiously optimistic," said opposition spokesman Aboubacar Sylla.

Political instability in Guinea following a military coup in December 2008 has deterred some investors, despite the country's large deposits of iron ore, bauxite, gold and other minerals.

Ravens hand out Super Bowl rings to champs

(AP) ? The Baltimore Ravens received their Super Bowl rings Friday night during a ceremony at the team's training complex.

Ray Lewis, who retired after the season, was among those in attendance. For Lewis, the ring will serve as a memento of his 17th NFL season.

Lewis said, "There's no better way to go out. That I can hold this the rest of my life and know I went out as champ."

The ring is made with white and yellow gold and the face is somewhat oval in shape. A purple Ravens logo encrusted in diamonds stands in the middle and is positioned over a pair of Lombardi Trophies. The Ravens won their other Super Bowl after the 2000 season.

There are 243 diamonds, the two largest atop the Lombardi trophies.

Andrea's rain spawns flood watches on East Coast

NEW YORK (AP) ? Soaking rains that spawned numerous flood watches pushed some streams and creeks over their banks throughout the Northeast, yet the first named storm of the Atlantic hurricane season sped up the Eastern Seaboard without causing major damage.

After bringing rain, strong winds and even tornadoes to Florida, Andrea lost most of its tropical characteristics late Friday into Saturday. The storm was packing winds that gusted between 20 to 30 mph, and officials warned of winds near gale force from New Jersey to Canada through Sunday.

The storm was blamed for one traffic-related death in Virginia.

Late Friday, the National Hurricane Center in Miami discontinued all tropical storm warnings, but cautioned about possible coastal and localized flooding from New Jersey to New England. As of 3 a.m. Saturday, the remnants of Tropical Storm Andrea were located near Cape May, N.J., headed northeast at about 35 mph (56 kph)

Dean Iovino, a meteorologist with the National Weather Service in Mount Laurel, N.J., said Saturday the storm was expected to pick up speed through the overnight hours, and could drop from 3 to 5 inches of rain in areas from New Jersey into New England before tapering off.

Officials in the Mid-Atlantic region and Northeast had prepared for the storm Friday night. New York City activated its flash flooding plan, and heavy rainfall resulted in a number of flash floods, causing some sections of roadways to be closed throughout Long Island.

A number of roads were flooded in the Boston area. A flight that left Boston on Friday night headed to Palm Beach was diverted to Newark International Airport in New Jersey after being struck by lightning. There were no injuries.

The weather service reported that small streams and creeks in southeastern Pennsylvania were going over their banks Friday night. New York City's airports experienced flight delays, and Connecticut reported numerous lane closures on highway as cars spun out amid heavy rain.

The rainy weather washed out events such as NASCAR's Sprint Cup qualifying in Pennsylvania's Poconos, and MLB games in Washington, New York and Boston.

The heavy rain from the storm's outer bands was being blamed by authorities in Virginia for a fatal accident on Interstate 77 in the state's western mountains. William Petty, 57, of Lexington, S.C., died when a car in which he was a passenger hydroplaned while passing a tractor-trailer. He survived the crash, only to be killed moments later when the car was struck by second tractor-trailer, authorities said.

During Friday's morning rush in Charleston, S.C., there was little evidence ? beyond a few downed tree branches, gusty winds and some puddles in the street ? that the center of the storm was passing nearby. The sun occasionally peeked through.

Derrec Becker with the South Carolina Emergency Management Division said the storm brought only a severe thunderstorm Friday. No injuries were reported, and there had been no reports of significant damage.

Mike Sprayberry of North Carolina Emergency Management told the Weather Channel that there had been some flash flooding and local road closures in the state but that "so far we have been quite fortunate."

Thousands of power outages were also reported.

Florida Gov. Rick Scott had warned Thursday of the risk of tornadoes, and officials said that eight were confirmed across the state.

There were no major problems along the most vulnerable parts of the coast such as North Carolina's Outer Banks.

David Tweedie, 41, of Ocracoke, said an early-morning burst of rain and the forecast of another three hours or so of rain and wind on the Outer Banks island has done little to alter the day's routine for the roughly 1,000 year-round residents.

The Friday fish fry that kicks off the island's annual folk music and arts festival was moved indoors to the island's only public school, and a musical performance of the three-day event was shifted to the community center. But the tropical system was otherwise forcing no changes to the Ocrafolk Festival that normally draws more than 2,000 visitors, Tweedie said.

Authorities in coastal Bertie County, N.C., said a school bus with 32 elementary students on board slid off the road and into a ditch about 8 a.m. No injuries were reported.

A Coast Guard cutter and HC-130 Hercules airplane were called to rescue four adults aboard a 35-foot sailboat about 65 miles off Charleston, S.C. The sailboat's engine was disabled during the storm and left rocking in 15-foot seas and 35-mph winds.

Farther east in Chapel Hill, heavy rains forced the forced the postponement of Friday's NCAA super regional baseball series opener between No. 1 seeded North Carolina and South Carolina until Saturday. A second game between N.C. State and Rice in Raleigh was also postponed.

While the storm departed Florida early Friday, the Sunshine State was still feeling the effects into the day. The weather service estimated that feeder bands from Andrea's remnants dropped more than 9 inches of rain on eastern Miami-Dade County and more than 6 inches of rain on eastern Broward County on Friday.

In Cuba, days of torrential rains associated with Andrea caused rivers to jump their banks in the western province of Pinar del Rio. More than 3,300 people evacuated endangered homes, and nearly 1,000 acres (4,000 hectares) of croplands suffered "serious damage," state-run newspaper Juventud Rebelde said Friday. Rain was forecast to continue falling on already waterlogged areas through Saturday.
